View Full Version : Cypripedium seed?
Jack April 3rd, 2007, 04:06 AM Hello,
I have sowed the following Cypripedien on Steel medium Octobers 2006.
reginae - my seed
passerinum - of Aaron Hicks
calceolus - of montanum
guttatum - of montanum
The first Protocorme are forming at the reginae now. Nothing still does himself at the other ones.
Does this always last for so long? Can I also expect germination at the other ones?
Best wishes
Jack
montanum April 3rd, 2007, 07:39 PM Hi Jack,
Sorry, often it does take this long! I think that the longest It's taken some of my seed to germinate is about 10-12 months. Dry Cyp calceolus is notoriously hard to germinate, which is why most people sow it green pod. I've had no luck with the guttatum seed either. Has anyone? Cyp passerinum is often tricky too!
Did you add potato to the medium?
Keep the flasks for a while. Bill Steele argues that if it hasn't germinated within a month or two, it won't. I know my technique isn't perfect, but personally, I have good germination up to 6-8 months out!
Best,
Ross
fundulopanchax April 3rd, 2007, 08:02 PM Be patient, sometimes it takes a long time! I have had flasks sit for a year with no germination at all then I look at it the next week and there are 500 protocorms.
If you have no protocorms at all you might want to place those flasks in the refrigerator for about 12 weeks then take them back out. Sometimes that will initiate mass germination.

Jack April 4th, 2007, 03:14 AM I then still can hope. I had potato in the medium and like you here have recommended bleached.
I have sowed which ones on 30-12-6 once again. I want 3 margins to fetch the 1 without Kühling, and the 2. 4 weeks cooling the 3. from the refrigerator now after approx. 12 weeks. I also do not have any results at the first two.
@montanum, Of the guttatum seed I have given an acquaintance something. This one had germination after 4-5 weeks. He used a modified Mamlgrenmedium with IAA and BA in the medium. I also will make once a seed at this medium now.
Best wishes

Loripep April 4th, 2007, 08:31 AM Hi there;
I had success with my germination the first time I tried guttatum (not blowing my own horn here, honest). they were bleached until that straw colour shown on one of Ron's posts. The media contained: 300mg charcoal, 0.2mg/L BA with 400mg/L casein and 400mg/L ammonium nitrate. Also used media without charcol and BA and a media -charcoal and 0.3mg/L BA. Growth was best on the first media.
Someone told me that soaking the seeds overnight in a tween solution at a pH of 12.3 heelps remove germination inhibitors but I haven't tried that yet.

Jack April 5th, 2007, 07:33 AM Hello,
I have talked to my acquaintance once again. This was his answer.
He took Malmgrenmedium modified without charcoal, with 0,2ppm BA. Be bleached with 2.5% CaOCL for 17 minutes. Germination approx. 30%.
Then the medium with 500 mg amino acids and 500 mg of charcoal without additions like pale ones. The germination is at about 30% there. "So one cannot say that one needs BA absolutely."
I then will make a seed with this medium, too and hope for similar results.
Best wishes

Jack April 23rd, 2007, 03:20 AM Hello Ross,
My guttatum are germinating now, too. I am happy.:) More Protocorm than at the reginae form there. Thought I rginae are easier than guttatum?
Best wishes

Jack April 27th, 2007, 03:40 AM How long do I have to leave the Protocorme on this medium?
Is the medium changed for the Protocorme? I used the Steele medium with potato.

If you use the Steele media without charcoal I find that it is good practice to reflask every two months or so - lots of toxic metabolites build up. If charcoal is added - anywhere from 400 mg/liter to 2 g/liter is used by different people, then the seedlings may be raised to time for vernalization. I would still recommend reflasking once however to thin the protocorms or the roots will become so tangled that many will be lost when the seedlings are harvested.

Jack April 28th, 2007, 03:01 AM Thanks Ron,
I did not take any charcoal. Is potato further used?
Best wishes

Hi, Jack,
I do continue to use potato - 1 cm cube per 100 ml of medium for everything except Cyp parviflorum - for them I use 3 cubes per 100 ml of medium.
