Hello all, I was wondering if anyone who has done chromosome squashes on paphs could tell me what method they use. I have seen a few types and procedures in orchid literature, but dunno which is optimal. Thanks

There is only one basic way to do it. Protocols may differ in the mitotic arrest chemical used; fixation, staining and whether the cover slip is removed or left in place but most of these differences depend on ones personal choice and experience. Chromosome work in plants is as much art as science and takes some experience to make it work. If you have some specific protocol questions or comparisons of specific points of protocols I will try to help.
